General description

Zinc finger protein with KRAB and SCAN domains 3 (UniProt: Q9BRR0; also known as Zinc finger and SCAN domain-containing protein 13, Zinc finger protein 306, Zinc finger protein 309, Zinc finger protein 47 homolog, Zf47, Zfp-47) is encoded by the ZKSCAN3 (also known as ZFP47, ZNF306, ZNF309, ZSCAN13) gene (Gene ID: 80317) in human. Zf47 is a member of the krueppel C2H2-type zinc-finger protein family. Two isoforms of Zf47 have been described that are produced by alternative splicing. It contains 7 C2H2 type zinc-fingers. It serves as a transcriptional factor that binds to the consensus sequence 5′-[GT][AG][AGT]GGGG-3′ and acts as a repressor of autophagy. It is shown to specifically repress expression of genes involved in autophagy and lysosome biogenesis/function such as MAP1LC3B, ULK1 or WIPI2. It is mainly localized in the nucleus, however, under conditions of starvation, it translocates to the cytoplasm, allowing expression of target genes involved in autophagy and lysosome biogenesis/function. Overexpression of Zf47 is observed in various tumors, including multiple myeloma, colorectal and prostate cancers.

Immunogen

KLH-conjugated linear peptide corresponding to 12 amino acids from the N-terminal half of human Zinc finger protein with KRAB and SCAN domains 3. The immunogen sequence is conserved in both isoforms, 1 and 2.
